Having done some research, the only thing a splatoon amiibo will do for this game, is unlock 15 single player challenges, where upon completion, you'll unlock some admittedly slick costumes. So no you don't need it, and there are plenty of very nice costumes already in the game. - You unlock a different set of gear for multiplayer. Girl unlocks a school uniform, boy unlocks Samurai gear, Squid unlocks mech armor.
- You unlock a new gun with each. Girl unlocks a charger, boy unlocks a roller, Squid unlocks a rifle.
- You unlock a new mini game with each one, though I'm not sure which mini game each gives. You also get a bonus minigame for completing all three sets of Amiibo challenges (overall 60 challenge levels). see less

In short - no real difference once you've got going. The data that makes up the game is the same on disc as it is via download - everything plays the same.
In detail:
Splatoon Disc: You have to insert the disc into the disc-drive of the Wii U in order to be able to play the game. You need to eject it again if you wish to play a different (disc) game. Some (but not much) storage space on the hard-drive (or additional storage unit) is needed for updates. The disc is vulnerable to physical damage if not looked after. If you look after the disc then you retain the option of selling the game on (or lending it to a friend) if you don't like it or have otherwise finished with it. The disc comes with a colourful (as pictured) case to look after it in. I like box art and collecting games so I have the disc version of Splatoon.
Download version: You can play the game very conveniently as you don't need to get off the sofa to find the disc, insert it etc. You need quite a lot of storage space for the full game, but if you're using a new 32gb Wii U then this won't be a problem. If you delete the game to make room for other games some time in the future, you'll be able to re-download the game for free (for as long as Nintendo maintains its availability). Because you're only getting the code, you won't be able to play the game 'immediately' - you'll have to get the Wii U connected to the internet and then go to the eshop to enter the download code and download and install the game. Depending on your internet speed this may take an hour or two. As there's no disc you can't damage it, and you can't sell it on in the future.
While I like having the game on disc I wish I had it installed on my hard-drive too as games like Splatoon are great for dipping in and out of. see less
